Rationale for the treatment of heart failure in patients with advanced heart failure: a review of pharmacokinetic, pharmacodynamics, and clinical data.
The pharmacokinetic parameters were evaluated in 3 separate trials, a total of 6 to 10 subjects in each trial, and the results showed that the mean AUC values were approximately equivalent within the range of 2–6%, and that the AUC values increased with dose (see Table ).
AUCs in the blood were approximately 3–5% higher after once-daily dosing compared with once-daily administration (see Table ).
In contrast, the AUCs in the liver were about 3% lower after once-daily dosing compared with once-daily administration (see Table ).
There were no statistically significant differences between the mean AUC values in the liver or blood in patients receiving Crestor and the placebo groups. AUC values were similar between Crestor and the placebo group, but in the liver, the mean AUC in the Crestor group was approximately 1.2% higher compared with the placebo group. There was no statistical significance with respect to the AUC values in the blood, AUC, or liver.
The AUC values in the blood were approximately 1.5% higher in the Crestor group compared with the placebo group, but the difference did not reach statistical significance (see Table ).
In the plasma, the mean AUC values in the Crestor group were approximately 1.3% higher compared with the placebo group, but the difference did not reach statistical significance (see Table ).
AUCs in the plasma were approximately 3.1% higher in the Crestor group compared with the placebo group, but the difference did not reach statistical significance (see Table ).
In the blood, the AUCs in the Crestor group were approximately 1.5% higher compared with the placebo group, but the difference did not reach statistical significance (see Table ).
In patients with mild to moderate (Child-Pugh class A or B), AUCs were approximately 2% higher in the Crestor group compared with the placebo group, but the difference did not reach statistical significance (see Table ).
AUCs were similar between the Crestor and placebo groups, but the difference did not reach statistical significance (see Table ).
The AUCs in the plasma in the Crestor group were approximately 2.1% higher compared with the placebo group, and the difference did not reach statistical significance (see Table ).
In the blood, AUCs in the Crestor group were approximately 1.3% higher compared with the placebo group, but the difference did not reach statistical significance (see Table ).
In patients with moderate to severe (Child-Pugh class B or C), AUCs were approximately 1.2% higher in the Crestor group compared with the placebo group, but the difference did not reach statistical significance (see Table ).
There were no statistically significant differences between the mean AUC values in the Crestor and placebo groups. AUCs in the liver were about 3.6% lower in the Crestor group compared with the placebo group, but the difference did not reach statistical significance (see Table ).
AUCs in the blood were approximately 3.1% higher in the Crestor group compared with the placebo group, but the difference did not reach statistical significance (see Table ).
In the plasma, the AUCs in the Crestor group were approximately 1.6% lower compared with the placebo group, but the difference did not reach statistical significance (see Table ).
The mean AUCs in the Crestor and placebo groups were approximately 1.4% higher in the Crestor group compared with the placebo group, but the difference did not reach statistical significance (see Table ).

AstraZeneca is to cut the cholesterol-lowering drug Crestor by 40 per cent on Tuesday, Health Canada said on Thursday.
The Canadian Health Products Regulatory Agency said the drug, which blocks the cholesterol-lowering drug, Crestor, will be removed from sale in the United States on Wednesday and other countries, including Australia and Canada, will also continue to sell it.
The drug's patent for the statin Merit Rx has expired in the United States, the FDA said in its notice of intent to halt distribution of the drug. The agency has also not yet decided whether to sell the drug in other countries.
The agency's notice of intent said the drug has been "discontinued" in the U. S. and Canada. It was not authorized for sale in Australia and Canada, it said.
It was "notifying the Canadian Food and Drug Administration (CFCA) and the Australian and New Zealand Medicines and Healthcare Products Regulatory Agency (ANPR), that Crestor was not being distributed in the U. S.," the notice said. It added that the drug's U. distributor has also not stopped making claims against the agency for the drug.
The agency said the drug had been under development since 2009.
The Canadian Health Products Regulatory Agency said it will begin finalizing the drug's approval in October.
Crestor, which is sold under the name Crestor, had been on the market since 2006. The drug has been under investigation by the FDA since the year 2009.
Crestor is one of several cholesterol-lowering drugs that are also used to treat high cholesterol and prevent heart disease.
